Abstract

Genetic variations among six barley genotypes were evaluated using Random Amplified Polymorphic DNA (RAPD) and Inter Simple Sequence Repeats (ISSR) markers. The six barley genotypes were highly different in their net blotch reaction (three genotypes Giza 126, Giza 2000 and Giza 132 were resistant, while the other genotypes namely; Giza 131, line 1 and line 2 were susceptible). Among these markers, RAPD primers generated 49 amplification products of which 14 were polymorphic, while ISSR markers produced 42 amplification products, out of which 22 were polymorphic. ISSR fingerprinting detected more polymorphic loci than the RAPD fingerprinting. The dendrograms constructed using RAPD and ISSR marker systems were not highly correlated with each other. However, RAPD draw clear distinguish among resistant and susceptible genotypes. Pairwise similarity index values ranged from 0.74 to 0.99 (RAPD) and from 0.65 to 0.82 (ISSR). RAPD and ISSR marker systems were found to be useful for the genetic diversity studies in barley but RAPD was more efficient to identify barley net blotch resistance.
