Abstract

Urease activity is often used for characterization of microbial viability in soil. The aim of the investigations was to measure the influence of chromium (III) on urease activity. Urease activity in pure solution was so sensitive for Cr3+, which caused inhibition of urease activity significantly. In this paper complexation reaction between Cr3+ and Jack bean urease is examined using isothermal titration calorimetry (ITC). It was found that chromium ions acted as a noncooperative inhibitor of JBU, and there is a set of 12 similar and independent binding sites for Cr3+ ions. The association equilibrium constant is 6.79×106 L-1.mol, indicating the strong interaction of Cr3+ ion with JBU. The molar enthalpy of binding is DH = 15.10 kJmol-1.
